This would depend! Mailer boxes are sturdy enough to be sent out on its own and are often used as external shipping cartons itself. If your mailer boxes have artwork on the outside of the box, you may want to add an external envelope/carton just to ensure your print is protected. Otherwise, you can ship it out as is!
What some brands do is add printing on the inside of the box only. They leave the outside of the box blank so there's room to place shipment labels, and when a customer opens the box they can see full branding on the inside of the box, which wouldn't be affected by the wear and tear of shipping.
